fMRI evaluation of NGF gene therapy on neural functional recoveryin the region of perihematoma after intracerebral hemorrhage

Abstract
Objective:To study the applied value of using fMRI to evaluate the therapeutic effect of NGF gene on neural functional re2covery in the region of perihematoma after experimental intracerebral hemorrhage (ICH).
Methods:Twenty2four healthy dogs were dividedinto three groups randomly:NGF group ( n= 12), ICH group ( n=8) and control group ( n=4). The examinations of Purdy score, MRscanning and TEM were carried out at 1, 3, 10, 28 day after ICH.
Results:Compared with ICH group, the volume of hematoma and ede2ma in NGF group had no significant difference ( P > 0.05), whereas ADC and rCBF in NGF group had significant difference at 1 - 3 day( P< 0.05). At 10 - 28 day, the volume of hematoma and edema, ADC and rCBF in NGF group were obviously better than that in ICHgroup ( P < 0.05). The dynamic changes of MR were correlated with the changes of clinic neural function and pathological result.
Conclur: sion fMRI was an objective and effective method to evaluate quantitatively and qualitatively the results of gene therapy.
